Research Fields

My research experience has been focusing on the male reproduction, especially on the accessory sex gland protein function in sperm activation and sperm membrane protein modification and sperm maturation. We had completed the transcriptome analysis of accessory sex gland and testis, and also the proteomic analysis of sperm, accessory sex gland and testis have been done. We had obtained such data set and want to find more interesting regulation mechanisms in spermatogenesis, sperm maturation, and interaction of spermatozoa and egg. Based on the high-throughput sequencing results and bioinformatics analysis we identified many important signal pathways involved in reproduction, such as MAPK and WNT signal pathways, I have applied four funds from National Natural Science Foundation of China and achieved perfect production in relevant research area. I studied in Cornell University as a visiting scholar for one year in 2014, and cooperation professor is Mariana Wolfner, who is member of the US National Academy of Sciences. She is excellent in reproduction research of Drosophila and I established the CRISPR/Cas9 system in her lab during visiting study and learned the accessory sex gland protein function research system. Male AGPs can also regulate proteolysis, thereby promoting the effective conduct of mating behavior. Serine protease proteins have been proved to play a role in this process in many species. Seminase causes protease to hydrolyze accessory gland protein in the process of starting the protease cascade signal pathway and participates in the early regulation process after mating. Serine protease 2 (Ser2) is also very important for reproduction in silkworm. We knockout the Ser2 gene using CRISPR/Cas9 technology in silkworm, it is found that Ser2 mutation causes sperm can’t enter the egg smoothly, resulting in male mutant sterility. Wild type females and Ser2 mutant males lay eggs normally but do not hatch after sexual mating, and Ser2 mutation does not affect the normal progress of other reproductive behaviors, Moreover, male sterility can be stably passed on to offspring. In addition, similar male sterility phenotypes were obtained by knocking out the Ser2 homologous gene in the lepidopteran pest Plutella xylostella. These results unanimously show that Ser2 gene is an essential factor for male reproductive success. It can be seen that proteases and protease inhibitors are another important protein family of paragonadal proteins that regulate sperm function.
