个人简介社会兼职研究方向1.微生物遗传育种 2.微生物代谢组学 3.微生物功能基因组学 招生与培养开授课程1. 微生物学及实验 2. 基因工程 3. 现代生物学大实验 4. 现代生物技术大实验 5. 生化与分子生物学原理与技术 科研项目自然基金: 1.国家自然科学基金青年项目,31100109,绿脓杆菌抑制抗菌肽HIP/PAP感染烧伤伤口的分子机制研究,2012/1-2014/12,22万元,已结题,主持 该项目确定绿脓杆菌模式识别受体通过相关信号通路来诱导人体角质细胞HIP/PAP表达及绿脓杆菌毒性因子又可以抑制人角质细胞HIP/PAP表达。构建小鼠烧伤模型,完成绿脓杆菌毒性因子调控小鼠角质细胞HIP/PAP表达实验;分离并鉴定调控HIP/PAP表达的绿脓杆菌毒性因子的组成,验证毒性因子对HIP/PAP的调节作用。我们阐明了绿脓杆菌的菌毛通过TLR2信号通路来诱导角质形成细胞HIP/PAP表达,抵抗细菌感染的分子机制。确定绿脓杆菌毒性因子抑制人角质细胞HIP/PAP表达实验 2.国家自然科学基金面上项目,31670138,绿脓杆菌致病性相关双组分系统的筛选及其作用机制研究,2017/1-2020/12,60万元,在研,主持 该项目通过Tnseq技术初步筛选到17个绿脓杆菌在皮肤感染中致病性相关的双组分系统基因,其中4个双组分系统基因功能未知。我们选择双组分系统基因PA14_46990构建了基因敲除菌株,从感染小鼠皮肤后的表型看,PA14_46990基因敲除菌株产生的皮肤坏死情况明显比野生型要轻,表现在坏死面积较小,同时分析感染后的感染部位绿脓杆菌载菌量也发现,PA14_46990基因敲除菌株的载菌量要比野生型菌株少。这些结果说明了PA14_46990基因在绿脓杆菌皮肤感染中起着重要作用,是一个致病性相关的双组分系统基因,接下来我们将对筛选到的双组分系统基因的调控绿脓杆菌毒性因子的分子机制及对宿主的炎症反应的应答机制作进一步的研究,以揭示绿脓杆菌双组分系统在皮肤感染中的重要作用。 学术成果代表性研究成果和学术奖励情况 一、期刊论文 Wei Zhang, Hongliang Gao, Yimin Huang, Shiqiang Wu, Jiangtao Tian, Yanning Niu, Chunjing Zou, Caifeng Jia, Mingfei Jin, Jing Huang, Zhongyi Chang, Xuexia Yang⁎, Deming Jiang⁎.
